Mackie stepped into acting with an understudy to Don Cheadle in Suzan-Lori Parks’ play Topdog/Underdog in 2002. The following year, he landed his role in Crossing and Hollywood Homicide.
With five movies, he came in 2004. Among them, Brother to Brother and She Hate Me led him to be nominated for many awards. On the same year, he landed his role in Million Dollar Baby which is one of his successful roles.
In 2005, he acted in The Man as Booty. The following year, Mackie acted in movies like Freedomland, Half Nelson, Heavens Fall, We Are Marshall and Crossover. He appeared on Ascension Day and Eagle Eye.

He acted in Captain America: The Winter Soldier in 2014. Again with the same role, he came up in Avengers: Age of Ultron. In the same year, Mackie appeared in Marvel’s Ant-Man.
With the role as Sam Wilson / Falcon, he again appeared in 2016’s Captain America: Civil War. In Avengers: Infinity War, he again made his appearance which was released in 2018.
