In 2004, she made theater her debut as Big Mama in Cat on a Hot Tin Roof. Also, she was nominated for Tomy Award for Best Featured Actress in a Play.

Along with that, she had worked in several Off-Broadway stage productions. Margo Martindale rose to the limelight after originating the role of Truvy Jones in the Steel Magnolias. Some of her Off-Broadway is Always…Patsy Cline and The Sugar Bean Sisters.
