Tennessee Titans

On May 14, 2018, Spillane signed as an undrafted free agent with the Tennessee Titans. On September 2, he re-signed to the Titans practice squad, he was made out from the 53-man roster at the end of training camp.

During a losing game against the Baltimore Ravens., he made his NFL debut, on October 14.

Pittsburgh Steelers

He signed the contract with the Pittsburgh Steelers on February 8, 2019. Before resigning to the contract on October 8, he was released from the contract on September 24, 2019. He ended his season by making 11 tackles in eight appearances.

He got injured tearing his ACL on October 18 2020 during the game against the Cleveland Browns. Ending the game with 5 tackles, he replaced Bush.

He was announced as the start for the remainder of the season by the Steelers coach Mike Tomlin, On October 20.  On March 24, 2021, he re-signed the one-year deal with the Steelers.

He ended his 2021 season with 56 total tackles in 14 appearances and four starts. Similarly, he scored one sack, 79 total tackles, and four passes defended in 16 games and five starts in the 2022 season.

Spillane was made a restricted free agent on March 11, 2022.
