George Wilson was first signed as an undrafted free agent by the Detroit Lions on April 30, 2004.

Around six months later, in October 2004, the Buffalo Bills signed him to their practice squad. He made his debut for the Bills on September 11, 2005, against the Houston Texans.

After that, George Wilson only made two more appearances till 2007. He made the transition to strong safety in 2007.

Unfortunately, his 2007-2008 season ended prematurely with a rib injury on December 10, 2007, against the Miami Dolphins.

In the next season, he featured in all 16 games of the season for the first time in his career.

On November 9, 2008, he shared a sack of the Patriots Matt Cassel with Marcus Stroud which marked his first career participation in a sack. Moreover, on the same play, he recovered the fumble forced by Stroud marking his second career fumble recovery.

At KC, he had four tackles, including one for a loss of five yards, recovered a fumble, and a sack.

With 15 special team tackles, he was voted special teams captain prior to the season and since then, he was voted consecutively as team captain.

Moreover, George Wilson became the first Buffalo Bills defensive back to record consecutive multi-sack seasons since Lawyer Milloy in 2003–04 being the second Bills defensive back to ever accomplish the feat.

The Bills released him on February 11, 2013. He is regarded as a Bills legend.

On February 22, 2013, the Tennessee Titans signed him on a two-year contract. Wilson retired with the Titans.
