DeSagana Diop was selected by Cleveland Cavaliers in the 2001 NBA draft. He played for the team Cavaliers till 2005 and then he signed with Dallas Mavericks. On 19 August 2005, he signed a three-year contract with the Mavericks. He recorded his first double-double season high with 10 points and 15 rebounds in a winning match against Minnesota Timberwolves. On 19 February 2008, he was traded to New Jersey Nets. On 9 July 2008, he resigned a six-year contract with the Mavericks worth $32 million but he was soon traded to Charlotte Bobcats on 16 January 2009. On 30 September 2013, he signed with the Cleveland Cavaliers but he was soon released on 25 October 2013.

On 11 November 2014, DeSagana Diop started as a player development coach for Texas Legend of NBA Development League and later he was promoted to assistant coach on 19 October 2015. On 3 October 2016, he was acquired by the Utah Jazz as a coaching associate.
