In 2001, he played as a defender for amateur club Cimiano from 2001 to 2002 at the time of his early career. He then went to play for the A.C Milan Youth Team in 2002. After the 2011 season was completed, he was promoted to play for the senior team. He played for the senior team until the 2011-17 season. With A.C Milan, he won 2 Supercoppa Italiana (2011, 2016). At that time, he was also selected for Serie A Team of the Year 2012-13 seasons.

On 20 July 2017, Mattia De Sciglio was sent to Juventus from 2017-20. During his seasons for Juventus, he made 44 appearances together. In Juventus, he won 3 consecutive Serie A-League titles (2017–18, 2018–19, 2019–20), 1 Coppa Italia (2017–18), and 1 Supercoppa Italiana (2018). Meret left Juventus and joined the French Team, Lyon on 5 October 2020, and is currently playing in the same team as a Full Back. His season-long contract with Lyon ends on June 30, 2021.

He also plays at the international level for the Italian National team. Sciglio made his senior international debut in March 2013. He along with his teammates won a bronze medal for the 2013 FIFA Confederations Cup following his participation in the 2014 FIFA World Cup and UEFA Euro 2016.
